Melanoma skin cancer remains incurable, and targeting heat shock protein 90 (Hsp90) is a promising therapeutic approach. In this study, we investigated the effects of 17-aminogeldanamycin, a potent inhibitor of Hsp90, the nuclear factor-kappa B (NF-kB) activity in a patient-derived melanoma cell lines and NRASQ61R BRAFV600E.

We did a time-lapse microscopy and flow cytometry to monitor changes in cell meeting and survival. NF-kB activity is determined by Immunodetection of phospho-p65 and assessment of gene expression of NF-kB dependent by quantitative real-time polymerase chain reaction (qRT-PCR), Western blotting and enzyme-linked immunosorbent assay (ELISA).

constitutive activity of p65 / NF-kB was evident in all melanoma cell lines. The difference in rates may be linked to genetic changes in CHUK, IL1B, MAP3K14, NFKBIE, RIPK1, and TLR4, while the difference in the level of gene transcripts-induced NF-kB revealed by PCR arrays that may arise from the contribution of other regulatory mechanisms. 17-Aminogeldanamycin markedly reduced levels of phospho-p65, but the level of total p65 protein is not changed, indicating that the 17-aminogeldanamycin inhibits activation of p65 / NF-kB.

This conclusion is supported by a significantly reduced expression of selected genes of NF-kB-dependent: cyclin D1 (CCND1), CXC motif chemokine ligand 8 (CXCL8), and vascular endothelial growth factor (VEGF), as shown in the level of transcript and protein, as well as the secretion of IL-8 and VEGF. Our studies show that 17-aminogeldanamycin can be used for efficient inhibition of NF-kB activity and a simultaneous decrease in the level of IL-8 and VEGF in melanoma extracellular environment.

BACKGROUND baicalin is isolated flavonoids from Scutellaria baicalensis roots and used in traditional Chinese medicine. Psoriasis is a persistent skin disease and chronic recurrent inflammation that is characterized by inflammation and increasing keratinocyte proliferation. This study aimed to investigate the effects of baicalin on HaCaT immortalized human keratinocytes in vitro and molecular mechanisms involved.

MATERIALS AND METHODS HaCaT keratinocytes cultured in increasing concentrations of baicalin at 6.25 pM, 12.5 pM, and 25 pM. Psoriasis established in vitro model using HaCaT cells were treated with tumor necrosis factor-alpha (TNF-alpha). MTT assay was used to asses cell viability and apoptosis.

Western blot was used to measure the expression of Bcl-2, Bax, pro-caspase-3 and cleaved caspase-3, and enzyme-linked immunosorbent assay (ELISA) is performed to detect inflammatory cytokines. Quantitative reverse transcription-polymerase chain reaction (qRT-PCR) was used to detect the level of STAT3 and p65 mRNA. RESULTS baicalin reduce cell viability and induces apoptosis of HaCaT human keratinocytes in a dose-dependent manner.

The increase in cell survival and expression of inflammatory cytokines by HaCaT cells induced by TNF-alpha was significantly inhibited by baicalin. Baicalin significantly inhibited the activation of STAT3 / NF-kappaB in HaCaT cells stimulated by TNF-alpha. CONCLUSION baicalin inhibits proliferation and inflammatory cytokine expression in HaCaT immortalized human keratinocytes in vitro by inhibition of STAT3 / NF-kappaB signaling pathway.
