Dexmedetomidine Post-Conditioning Alleviates Cerebral Ischemia-Reperfusion Injury in Rats by Inhibiting High Mobility Group Protein BBNuclear Factor kappa B (NF-κB) Signaling Pathway.
BACKGROUND Cerebral ischemia-reperfusion injury is an important cause of death due to cerebrovascular accidents. increased research effort is needed to unravel the underlying mechanism of aggravation or reduction. In this study, the effects of dexmedetomidine post-conditioning in the HMGB1 / TLR4 / NF-kappaB signaling pathway in rat brain ischemia-reperfusion explored.
MATERIALS AND METHODS Ninety rats were randomly divided into 5 groups – groups of sham (Sham), a model group (I / R), a group of dexmedetomidine post-conditioning (Dex), high mobility group protein group B1 recombinant (rHMGB1), and HMGB1 + dexmedetomidine recombinant post-conditioning group (rHMGB1 + Dex) – with 18 rats in each group.
Longa levels, wet-dry weighing, TTC staining, HE staining and immunohistochemical staining was used to assess brain damage. ELISA, RT-PCR and Western blot analysis was performed to assess the expression of IL-1SS, TNF-alpha, IL-6, IL-8, HMGB1, TLR4 and NF-kappaB. RESULTS Compared with group I / R, neurological function score, the water content of the brain, the area of infarction, and the amount of COX-2 and IBA-1-positive cells in the group Dex significantly lower, accompanied by a decline in the function of HMGB1 / TLR4 / NF-kappaB pathway, alleviated inflammation and oxidative stress injury in brain tissue.
This trend reversed in the group mostly rHMGB1 and rHMGB1 + Dex group, but not in the Dex group. Furthermore, when compared with the group Dex, there was a significant increase of H₂O₂, MDA, NO, IL-1SS, TNF-alpha, IL-6, IL-8, HMGB1, TLR4, and p-P65 in the group rHMGB1 and rHMGB1 group + Dex, where a significant reduction of T-AOC, SOD, and p-IkappaBalpha also detected.
CONCLUSIONS Dexmedetomidine post-conditioning can relieve ischemia-reperfusion injury in rat brain by inhibiting pathways HMGB1 / TLR4 / NF-kappaB signaling.

BACKGROUND Microglia stay in the spinal cord plays a key role in the onset, the development of post-spinal cord injury (SCI) neuroinflammation. Curcumin has been shown to exhibit a variety of anti-inflammatory activity and anti-tumor. The purpose of this study was to explore the effects of curcumin on inflammatory responses in lipopolysaccharide (LPS) -activated microglia and mechanisms.
MATERIALS AND METHODS The expression levels of phosphorylated-p65 (p-p65), tumor necrosis factor (TNF) -alpha, interleukin (IL) -1ß, and IkappaB kinase ß (IKKß) is checked by western blot assay. Expression of MiR-199b-5p detected by real-time quantitative polymerase chain reaction assay. Suspected binding site of miR-199b-5p in the 3’UTR IKKß predicted by bioinformatics, and direct interaction between miR-199b-5p and IKKß verified by dual-luciferase reporter assay and RNA-immunoprecipitation test.
RESULTS Curcumin significantly suppressed the inflammatory response induced by LPS with inactivation of nuclear factor kappa B (NF-kappaB) in microglial cells, which reflected a decrease in the level of p-p65, as well as pro-inflammatory mediators, including nitric oxide inducible nitric (iNOS), TNF-alpha, and IL-1SS. Additionally, curcumin increased the level of miR-199b-5p and decreased expression of activated microglia cells IKKß in.
Knockdown of miR-199b-5p or excess of IKKß reverse the inhibitory effect of curcumin on inflammatory responses and activation of NF-kappaB. MiR-199b-5p directly targeted IKKß and repressed expression. IKKß abolished silencing of miR-199b-5p-stimulate the production of inflammatory cytokines and activation of NF-kappaB.
