BACKGROUND Compound pig cerebroside and ganglioside injection (CPCGI) has been widely applied in clinical practice in China for treating functional confusion caused by brain disease. Sevoflurane, the inhalation anesthetics are often used, was found to have neurotoxicity which can cause nerve damage in patients. This study was conducted to investigate the protective effect of CPCGI on sevoflurane-induced nerve damage and to express the neuroprotective mechanism of CPCGI.
MATERIALS AND METHODS First, hippocampal neurons are separated from Sprague-Dawley rat embryos, and stimulated by 3% sevoflurane for different times (0, 2, 4, and 6 hours). Then, the cell viability and apoptosis assessed by thiazolyl blue tetrazolium bromide (MTT) and cytometry (FCM) flows, respectively. Western blot analysis was used to determine the expression levels of proteins associated with apoptosis.
RESULTS The results showed that 3% sevoflurane significantly inhibited cell viability but induced apoptosis in neurons with a time-dependent manner. Treatment with 3% sevoflurane also promoting Bax (B cell leukemia / lymphoma 2 (Bcl-2) -associated X protein) and caspase3 cleaved protein expression, and suppressed the expression of Bcl-2 and pro-caspase3 in hippocampal neurons.
In addition, phosphorylated (p) -p38 and the expression of p-p65 and the ratio of p-p38 / p38 and p-p65 / p65 are regulated depending on time after 3% sevoflurane treatment. Further analysis showed that all the effects of 3% sevoflurane in reverse hippocampal neuron with a pre-treatment CPCGI.
CONCLUSION We showed nerves of CPCGI role in sevoflurane-induced nerve cell damage through the Regulation of MAPK / NF-kappaB signaling pathway.

Grilled Nux Vomica influence on RNA Differential Expression Profile gastrocnemius muscle and Toll-Like Receptor 4 (TLR-4) / Nuclear Factor kappa B (NF-kB) Signaling in Rat Experimental Autoimmune Myasthenia Gravis.
BACKGROUND Myasthenia gravis (MG) is a progressive autoimmune disorder caused by the production of antibodies directed against acetylcholine receptors (AChRs), resulting in muscle weakness and fatigue. This study aimed to explore the effects and mechanism of grilled nux vomica (GNV) in experimental autoimmune myasthenia gravis (EAMG) mice.
MATERIALS AND METHODS Mice 97-116 peptides used to mediate the disease in the model EAMG in SPF female Lewis rats. The treatment group received grilled Nux Vomica (75 mg / kg, 150 mg / kg, and 225 mg / kg). The autoantibodies and inflammatory cytokine levels were measured by enzyme-linked immunosorbent assay (ELISA). RNA profiles done on the high dose group and rat models. Results profiling and TLR-4 / NF-kappaB signaling is validated by q-PCR and Western blot analysis.
RESULTS The results showed that GNV can attenuate symptoms EAMG mice. There are levels of AChR-ab, IFN-γ, TNF-alpha, IL-2, IL-4 and IL-17 levels decreased, and increased levels of TGF-ß1. In total, 235 genes differentially expressed (degs), which consists of 175 degs 60 degs upregulated and downregulated, are identified. functional annotation indicates that degs mostly associated with cell adhesion of leukocytes cells, NF-kappa B signaling pathway, muscle contraction, and cardiac muscle contraction track.
Rac2, Itgb2, Lcp2, Myl3, and Tnni1 regarded as the hub genes with a higher level of value in protein-protein interaction (PPI) networks. Q-PCR and Western blot results are consistent with the profile of the hub genes RNA. GNV treatment also significantly reduced NF-kappaB p65 protein expression of TLR-4 and in mice EAMG.

Conclusions These results suggest that ameliorates EAMG Grilled Nux Vomica by pressing the TLR-4 pathway / NF-kappaB signaling, and gene hub can serve as a potential target for the treatment of MG.
