Efficacy of green tea, its polyphenols and nanoformulation in experimental colitis and the role of non-canonical and canonical nuclear factor kappa beta (NF-kB) pathway: a preclinical in-vivo and in-silico exploratory study
NF-kB plays a major role in aetiopathogenesis of-inflammatory colitis. In this study, we evaluated the efficacy of green tea and polyphenols and their nanoformulation Tri-Nitro Benzene Sulfonic acid (TNBS) induced colitis in in-vivo systems (Rat) and the involvement of non-canonical and canonical NF-kB green line (platform di- silico) protection mediated tea. We use Wister rat model of TNBS-induced colitis.
Rats are grouped into eleven groups (six animals each) and administered vehicle (ethanol), TNBS, Epicatechin (EC), epigallocatechin (EGC), Epicatechin-gallate (ECG), epigallocatechin-gallate (EGCG), sulfasalazine, green tea, EGCG + sulfasalazine, nano-nano-EGCG and EGCG + sulfasalazine for 14 days after the induction of colitis. colonic tissue were evaluated for levels of malondialdehyde, myeloperoxidase activity, catalase, reduced glutathione, glutathione peroxidase, IL-6, TNF-α, IL-1β, NF-kB and morphological and histopathological evidence of damage.
In part, in silico, molecular docking and EGCG dynamic simulation studies carried out on different targets in the NF-kB for a detailed evaluation of the role of non-canonical and canonical NF-KB pathway. In our study, EGCG reduces colon inflammation, oxidative stress markers, TNF-α, NF-kB, IL-1β and IL-6. Nano-EGCG + sulfasalazine is more efficacious when compared to EGCG + sulfasalazine.
In molecular docking and molecular dynamics simulation studies, EGCG showed good binding sites profile by IKK-beta inhibitors, IKK-alpha and NIK binding. Thus, it can be concluded that EGCG showed protective action in experimental colitis acting through both canonical and non-canonical NF-kB pathway. Nano-EGCG + sulfasalazine combination showed a better protection than the nano-EGCG alone. Communicated by H. Ramaswamy Sarma.

Retinoic acid-Platinum (II) Complexes [RT-Pt (II)] Protect Against Rheumatoid Arthritis in Mice via MEK / Nuclear Factor kappa B (NF-kB) Pathway Downregulation
BACKGROUND Rheumatoid arthritis (RA) is an inflammatory disorder that is present in approximately 1% of the world population. This study aims to determine the effect of retinoic acid-platinum (II) complex [RT-Pt (II)] in rheumatoid arthritis (RA) and to explore the mechanisms involved.
MATERIALS AND METHODS MH7A cell viability was determined by MTT assay and apoptosis was assessed using a FACSCalibur flow cytometry. RT-PCR and Western blot tests used to assess the mRNA and protein levels.
RESULTS Treatment of rheumatoid arthritis with RT-Pt (II) significantly reduced levels of IL-1SS, IL-6, IL-8, MMP-1 and MMP-13 in the synovial fluid of mice with a dose-dependent manner. Expression of iNOS and COX-2 mRNA and protein in rats with rheumatoid arthritis also significantly inhibited by treatment with RT-Pt (II). proliferation of TNF-alpha-induced cell MH7A was overcome by RT-Pt (II) treatment with a concentration-dependent manner.
In addition, RT-Pt (II) treatment induced apoptosis and causing cell cycle arrest in cells MH7A. Activation of MEK / NF-kappaB pathway were downregulated by RT-Pt (II) MH7A cell treatment. CONCLUSION In summary, this study showed that the RT-Pt (II) inhibit TNF-alpha-induced inflammatory response, suppress cell survival, and induces apoptosis in synovial cells of rheumatoid arthritis.

In addition, RT-Pt (II) shows the effect by targeting the MEK pathway / NF-kappaB. Therefore, RT-Pt (II) can be used for the development of treatments for rheumatoid arthritis.
